The active constituents of the plant are mesembrine-alkaloids along with the closely related compounds of mesembrenone, mesembrenol and tortuosamine, which have been demonstrated by the National Institute of Mental Health (USA) to act positively on serotonin, nor-adrenaline, nicotine and dopamine receptor-sites in the brain.
As a relaxant Sceletium tortuosum (more commonly known as Sceletium) is unsurpassed, the first written record of it being "prescribed" is in the 1600's but reputedly it has been known in its primary form since man first walked the earth! It is still incomparable as a safe and unrivaled relaxation product for increasing relaxation, decreasing tension and managing apprehension.

The active ingredients from the primary plant used are alkaloids, including mesembrine, mesembrenol, mesembrenone, and tortuosamine. The alkaloid concentration in the dry material ranges from 0.05 to 2.3%. Mesembrine is usually the major alkaloid present, and has been demonstrated in laboratory studies (sponsored by the National Institute of Mental Health in the United States, and conducted by the company NovaScreen) to be a very potent serotonin-uptake inhibitor.
